Abstract: Objective To develope a method for staining Nissl bodies and myelin sheath in spinal cord tissues and the spinal ganglion from 8 New Zealand white rabbits. Methods The double-compound staining dyes composed of brilliant cresyl violet, toluidine blue,thionin,orange G and phosphotungstic acid (BTT-OP method) were investigated. Results The Nissl body,axons, nerve membrane and connective tissue were stained blue, the nerve cell nuclei were stained yellow, the myelin sheath and the background showed slight yellow. Conclusion Double-compound staining method (BTT-OP method )possesses more vivid color and more striking contrast than the original method, it can demonstrate clearly the structures of neuron under the light
